A Small-scale Northern Hybridization with Digoxigenin- labeled RNA Probes from PCR-based Templates

چکیده

CONTENTS Abstract We describe the use of PCR to generate sense and antisense template DNA for nonradioactive in vitro transcription reactions. The resulting digoxigenin-labeled RNA probes can be used in a small-scale Northern hybridization procedure that is performed in microcentrifuge tubes. This kind of Northern approach is especially useful when the expression of several genes is to be initially checked, for example, with a set of cDNA clones from a library screening.

Hybridization and detection of digoxigenin probes on RNA blots.

This article describes nonradioactive probing of a Northern blot. The method employs digoxigenin-labeled probes. Antidigoxigenin antibody/alkaline phosphate conjugate, and a chemiluminescent substrate are subsequently used in the detection system.

Non-radioactive labeling of RNA transcripts in vitro with the hapten digoxigenin (DIG); hybridization and ELISA-based detection.

We have developed a system for the enzymatic in vitro synthesis of non-radioactively labeled RNA which is derivatized with the hapten digoxigenin (DIG). The labeling reaction as well as the conditions for hybridization and detection of hybrids by an antibody-conjugate and a coupled colour reaction were analyzed and adapted for high sensitivity and low background. In addition, data on the perfor...

Identification of vaccine-related polioviruses by hybridization with specific RNA probes.

We developed RNA probes for the identification of poliovirus isolates by blot hybridization. Two sets of vaccine strain-specific probes were prepared. They complemented variable genomic domains within (i) the 5'-untranslated region and (ii) the amino-terminal codons of VP1. An enterovirus group probe (EV/5UT) matching highly conserved 5'-untranslated region sequences was used to estimate the qu...

An optimized protocol for high-throughput in situ hybridization of zebra finch brain.

In situ hybridization (ISH) is a sensitive technique for documenting the tissue distribution of mRNAs. Advanced nonradioactive ISH methods that are based on the use of digoxigenin (DIG)-labeled probes and chromogenic detection have better spatial resolution than emulsion autoradiography techniques and, when paired with high-resolution digital imaging, allow for large-scale profiling of gene exp...

Sensitive nonradioactive dot blot/ribonuclease protection assay for quantitative determination of mRNA.

We have developed a simple and sensitive method for the rapid quantitation of mRNA from cell cultures and small tissue samples. The method combines the high sensitivity and specificity of the ribonuclease protection assay with simple handling and rapid execution of dot blotting. The use of digoxygenin-labeled cRNA probes eliminates all problems associated with radioisotopes commonly used in the...
